Unique bacterial communities and lifestyles in deep ocean blue holes: Insights from the Yongle Blue Hole (South China Sea)

Deep ocean blue holes possess steep physicochemical gradients, especially low dissolved oxygen concentration, which shape the extraordinary microbial communities. However, environmental responses of microorganisms with different lifestyles and knowledge culturable in are still unknown. Here, we investigated bacterial community structure world’s deepest hole - Yongle Blue Hole (YBH) South China Sea using both culture-dependent -independent methods. YBH can be divided by content into an oxic zone, a suboxic zone two anoxic zones. The abundance bacteria, archaea, genes dsrB soxB were all higher free-living (FL) lifestyle than particle-associated (PA) lifestyle, yet diversity richness PA bacteria that FL bacteria. More Gammaproteobacteria less Alphaproteobacteria , Chloroflexi Nitrospinae observed within fraction fraction. relative sulfur-oxidizing (SOB) was dominant between 100-140 m (anoxic I) YBH, maximum 90.0% (140 fraction). SOB mainly colorless sulfur purple non-sulfur Thiomicrorhabdus Sulfurimonas main representatives. In addition, total 294 strains isolated on variety media culture conditions, 22.2% (18/81) anaerobic identified as potential novel species. Our study reveals distinction YBH. It contributes to further understanding deep holes, provides resources for subsequent studies their adaptation extreme marine environments.
